If you have done what Alex mentioned above and what I noted in post 5 about restarting RW's after you loaded the marker files, selecting route markers radio button on the bottom left flyout and still have issues I suggest you remake the files.
It only takes a few minutes as you already know and this will narrow it down to the .csv file if it still doesn't work. You should still have your .csv file in your Source Folder.
First up, delete these files in your Assets\Micheal\Addon03\ RouteMarkers folder.

GPSM Arker set blueprint001.bin
GPSM Arker set blueprint001.bin.tgt
GPSM Arker set blueprint001.xml
GPSM Arker set blueprint001.xml.tgt Then delete the old ******.xml blueprint file for your markers in your Source\Micheal\Addon03\RouteMarkers folder. You should have the .csv file still there.
I renamed this .csv file to Hood River for an example.
Okay, fire up RailWorks, clear cache and then launch the Blueprint Editor. Open up your Source folder to the RouteMarkers...you will see the .csv file I renamed to Hood River. Right click on RouteMarkers, choose the Blueprint and okay.

Rename the Blueprint to Hood River.

Blueprint is Hood River as done in prior picture, Type Hood River in English slot, move sliders as shown....the green should be 1, didn't move it far enough, type in RouteMarkers\Hood River.csv under series marker file tab and when happy click the export button. You should see a successful build at the very bottom when done.

Go to your Assets\Micheal\Addon03\ RouteMarkers folder and you should have these files. You can delete the three .tgt files.

Fire up RailWorks again, check the Route Markes radio button in the bottom left flyout in the WE save and exit. Start RailWorks up again, check the radio button is still active and check your cyan series markers.
If they are still not there here's a couple of things to check:
1. The long values ( -*** ) are in the left column of your csv file
2. The start long & lat are the same value in your Route Blueprint as what you used in ReDem.
