I think, or at least I find, that the Siding and Platform markers are more or less permanent with the route, where the Destination markers are used with scenarios. Think about it, sidings and platforms usually don't change in the route, where Destination markers tell the engineer where to go in a scenario. Destination markers can also be permanent, but I relate them to scenarios. So, I all reality, all the markers can be permanent, but the Destination markers are more related to scenarios than the others.
By modifying the marker's local position you change nothing but the end handles. Also, when placing the markers you can just position them where you want to start, click, hold, and slide to the end of the area of track you want to mark then release.
Just adding my two cents, I'm no expert, and will never claim to be.
